Citrus Compound Shows Promise Against Obesity

Scientists have found that a natural substance in some citrus fruits may help protect against obesity and health problems linked to it.

The compound, called nobiletin, is found in fruits including sweet oranges and tangerines.

Researchers at Western University in Canada studied nobiletin because obesity is closely connected with several major diseases. Carrying too much body fat can increase the risk of type 2 diabetes, heart disease, fatty liver disease and other long-term health problems.

The research team tested the compound in mice that were fed a diet high in fat and cholesterol. This type of diet is often used in laboratory research to study weight gain and changes in the way the body processes fats and sugar.

Some of the mice also received nobiletin. Despite eating the unhealthy diet, these mice remained noticeably leaner than mice that did not receive the citrus compound.

They also showed less insulin resistance. Insulin is a hormone that helps move sugar from the blood into cells, and insulin resistance develops when the body’s cells stop responding properly to it.

Over time, insulin resistance can cause blood sugar levels to rise and is an important step in the development of type 2 diabetes. The mice given nobiletin also had healthier levels of fats circulating in their blood.

The researchers then examined whether nobiletin could help after obesity-related problems had already developed. Their results suggested that the compound could reduce some of these harmful changes rather than only preventing them from appearing.

One particularly important finding involved atherosclerosis. This condition develops when fatty material and other substances build up inside arteries, forming plaques that can narrow the blood vessels.

Atherosclerosis is a major cause of heart attacks and strokes. In the study, nobiletin reduced plaque buildup in the arteries of mice, suggesting that its effects might extend beyond weight control.

The scientists also wanted to understand how nobiletin produced these changes. They initially focused on an enzyme known as AMP-activated protein kinase, or AMPK, which acts as an important energy sensor inside cells.

AMPK can encourage the body to burn stored fat and reduce the production of new fat. Because of these effects, scientists thought nobiletin might work by activating this system.

Surprisingly, experiments showed that nobiletin could still produce beneficial effects without depending on AMPK. This suggests that the citrus compound acts through another biological pathway that researchers have not yet fully identified.

Understanding that pathway could be important if scientists eventually try to turn the discovery into a treatment. It may also help researchers understand whether nobiletin could be combined safely and effectively with medicines that influence other metabolic pathways.

However, the findings should not be interpreted to mean that eating oranges or tangerines will prevent obesity or cure diabetes. The research was conducted in mice, and the amount and form of nobiletin used experimentally cannot simply be compared with eating ordinary citrus fruit.

Animal studies are useful for discovering possible treatments, but substances that work well in mice do not always produce the same results in humans. Human clinical trials would be needed to determine an effective dose, possible side effects and whether nobiletin actually improves metabolic or heart health in people.

The study was conducted by Murray Huff and colleagues at Western University and published in the Journal of Lipid Research. The findings add to growing scientific interest in natural plant compounds as possible starting points for future treatments for obesity, diabetes and cardiovascular disease.
