
Alzheimer’s disease develops slowly over many years before memory problems become obvious.
Scientists have long known that poor sleep and Alzheimer’s risk are connected, but a new study from Edith Cowan University suggests the picture is much more complex. Your genes may determine how strongly your brain is affected by sleep problems.
The research was completed by ECU’s Centre for Precision Health and published online in Alzheimer’s & Dementia, the Journal of the Alzheimer’s Association.
The team investigated a gene called AQP4, which plays a key role in moving fluid through the brain and supporting its natural waste removal system.
During deep sleep, the brain becomes especially active at clearing away unwanted materials. Researchers believe this nightly cleaning process removes proteins that can build up in Alzheimer’s disease.
If sleep is disturbed for many years, this protective system may not work as efficiently.
The scientists compared different AQP4 gene variants with participants’ reported sleep patterns, brain scans, and cognitive test results. Instead of finding one simple answer, they discovered that the effects of poor sleep depended on which version of the gene each person inherited.
For some people, shorter sleep was linked to faster shrinking of grey matter. Others were more affected by taking a long time to fall asleep, with scans showing lower brain volume. Thinking and memory also changed differently across the different genetic groups.
These findings suggest that not everyone responds to poor sleep in the same way. Two people could have similar ages, lifestyles, and medical histories but experience different rates of brain aging because their genes influence how the brain responds to sleep loss.
The researchers believe this knowledge could eventually help doctors design more personalized prevention plans. Instead of giving identical advice to everyone, future care might identify who would benefit most from intensive sleep improvement programs. However, the scientists say the evidence is still too early to recommend genetic testing.
Sleep remains one of the few Alzheimer’s risk factors that many people can improve. Maintaining regular sleeping hours, treating sleep disorders, reducing stress, and following healthy daily habits may support brain function throughout life.
More research will be needed to confirm the results in larger groups of people from different backgrounds. Clinical trials will also need to test whether improving sleep can reduce the extra risk linked to certain genetic variants.
