
Could the quality of your sleep influence your brain health decades before memory problems appear?
A new study from Edith Cowan University (ECU) suggests the answer may depend not only on how well you sleep but also on your genes. The research offers fresh clues about why some people may be more vulnerable to Alzheimer’s disease than others.
The study was carried out by researchers from ECU’s Centre for Precision Health and was published online in Alzheimer’s & Dementia, the Journal of the Alzheimer’s Association.
The scientists focused on a gene called aquaporin-4, or AQP4, which helps control the movement of fluid through the brain.
Scientists believe the brain has a natural cleaning system that becomes much more active during sleep.
This system helps remove waste products, including proteins that have been linked to Alzheimer’s disease. When sleep is poor or too short, this cleaning process may become less effective over time.
The researchers wanted to know whether different versions of the AQP4 gene change the way sleep affects the brain. To answer this question, they examined 13 common forms of the gene together with participants’ sleep habits, brain scans, and thinking and memory test results.
They found that the relationship between sleep and brain health was not the same for everyone. People carrying certain AQP4 variants showed faster loss of grey matter when they regularly slept for fewer hours. Grey matter contains many of the brain cells responsible for memory, learning, movement, and decision-making.
In some participants, taking longer to fall asleep was also linked with lower brain volume. Changes in thinking ability over time were different depending on both sleep quality and the person’s genetic variant. This suggests that genes and lifestyle work together rather than acting separately.
The findings may help explain why two people with similar overall Alzheimer’s risk can experience very different rates of brain aging. One person may be more affected by poor sleep because of their genetic makeup, while another may be more resistant.
The researchers stressed that the results do not mean people should seek genetic testing today. More studies involving larger and more diverse populations are needed before the findings can be used in everyday medical care. However, the study highlights sleep as one of the few risk factors people can improve.
Good sleep habits include keeping a regular bedtime, limiting caffeine late in the day, staying physically active, and creating a quiet sleeping environment. These healthy habits support overall wellbeing and may also help protect the brain as people age.
The research points toward a future where Alzheimer’s prevention may become more personalized instead of using the same advice for everyone. Review and analysis: This study is important because it combines genetics with lifestyle rather than treating them as separate issues.
Although it cannot prove that improving sleep will prevent Alzheimer’s, it provides strong reasons to test this idea in future clinical trials and shows why healthy sleep remains an important part of brain health.
