Natural Plant Compound Could Fight Rheumatoid Arthritis

Rheumatoid arthritis is a long-term autoimmune disease in which the immune system mistakenly attacks healthy joints.

This causes pain, swelling, stiffness, and gradual damage that can make everyday activities difficult. Although modern medicines help many people, some patients do not respond well or develop unwanted side effects, creating a need for better treatments.

A new study published in the journal Engineering suggests that a natural compound called obakulactone could become a promising new option in the future. Obakulactone, often shortened to OL, is extracted from Phellodendri cortex, a medicinal plant that has been used in traditional herbal medicine for many years.

The research team wanted to understand exactly how OL works inside the body. Instead of simply reducing inflammation, they found that the compound acts on important biological pathways that control immune activity, fat metabolism, and the survival of inflammatory cells.

The scientists tested OL in rats with rheumatoid arthritis. The animals received different doses of the compound for three weeks. Compared with untreated animals, the treated rats developed much less joint swelling and showed healthier cartilage and joint lining tissues under the microscope.

The compound also improved changes in the spleen and thymus, two organs that play important roles in the immune system. Inside the joints, OL reduced inflammatory immune cells while increasing cells that help calm inflammation and support healing.

Blood tests showed lower levels of several inflammatory substances that are commonly found in people with rheumatoid arthritis. The treatment also reduced important disease markers that doctors often use to monitor the condition.

Using advanced laboratory techniques, the researchers discovered that rheumatoid arthritis disrupted the body’s normal handling of healthy unsaturated fatty acids. OL helped restore a more normal balance, suggesting that correcting fat metabolism may be an important part of treating autoimmune disease.

The team also found that OL directly attached to a protein called ACOT1. Once attached, the compound encouraged cells to break down this protein naturally. As ACOT1 levels fell, several inflammation-promoting signaling pathways became much less active, reducing the growth and harmful behavior of joint cells.

This discovery identifies ACOT1 as a possible new drug target for rheumatoid arthritis. Future medicines based on this mechanism may control disease differently from current treatments and could potentially help patients who do not respond to existing drugs.

This research provides encouraging early evidence that obakulactone may reduce inflammation and joint damage by targeting fatty acid metabolism and a protein called ACOT1.

However, the work was carried out in rats and laboratory cells, so it cannot show that the treatment will work in people. Clinical studies will be needed before obakulactone can be considered as a new treatment for rheumatoid arthritis.
